Mechanism of action

Long-acting amylin analog (the short-acting analog is pramlintide / Symlin). Binds amylin and calcitonin receptors to delay gastric emptying, suppress glucagon, and produce satiety. Co-formulated with semaglutide as CagriSema; combined effect is additive on weight loss.

Common impurities & failure modes

  • Linear (uncyclized) precursor

    +2 Da vs. main peak; the most important impurity to quantify on cyclic peptides.

  • Misfolded disulfide isomers

    When other Cys residues are present, scrambled disulfides can form. LC-MS with non-reducing conditions resolves.

  • Aggregates

    Common in lipidated amylin analogs.
